"One" is not a number.

On a foggy night, a boat carrying a coffin was floating on the ocean. A female vampire, who wore a golden choker around her neck, rose from that coffin as soon as she ran aground on Duel Academy Island.

Meanwhile, resting and recuperating in the school infirmary, Jaden and Atticus—formerly known as Nightshroud—are still weak from the first Shadow Duel, while the other Key Keepers are informed by Chancellor Sheppard to keep an eye out for anything suspicious, as rumors of an invading vampire on the island had spread fast. However, as they prepared their Decks, the Key Keepers did not realize that eavesdroppers were eyeing their Decks—a group of bats summoned by the rumored vampire.

The Key Keepers, minus Jaden and Alexis—who'd stayed behind to look after her brother—made their way to the shoreline, where a blood-red carpet rolled out before them. Moments later, approaching them on the carpet was a beautiful but evil-looking woman, who identified herself as Camula, and she offered a challenge to the Key Keepers. As her eyes fell on Zane Truesdale, Dr. Crowler—after being encouraged to accept, due to his inflated ego—reluctantly volunteered to duel the vampire.

However, Camula's Vampire-like Zombies soon back Crowler into a corner and leave him stuck there, and before too long, Crowler's soul is sealed away in a small doll, while Camula is able to open one of the Spirit Gates holding the Sacred Beasts. Is this the beginning of the end for the world?
